日本を始め、多くの先進国は超高齢社会を迎えつつあり、老人に多い脳神経疾患の一つであるアルツハイマー病の治療薬が遍く待望されている。そこで、本研究はその原因タンパク質Aβ42 に特異的に結合し、重合を阻害するペプチドの開発を目指した。本年度の研究では先行研究(「地域結集型埼玉バイオプロジェクト」での研究)を受け、Aβ42 に結合するペプチドとして予備的にスクリーニングされたペプチド群の解析を行ない、評価系の開拓を行うとともに、新たな分子ライブラリー(“環状化可能ペプチド”など)を構築し、一層有力な分子をスクリーニングする準備を行った。この際、迅速スクリーニングのために体積活用型マイクロアレイ(MMV)の実用化を進め、反応条件・操作法の改良に努meた。 一方、周知の如く、高齢社会では「ガン治療」が、重要な医学テーマである。そこで、先 行研究からガン抑制効果が期待されている「カテプシンE“活性向上”ペプチド」の淘汰を すすめ、高い活性を有する数種のペプチドを得た。この際に、ペプチドーペプチド相互作用 を記述するために有効な「タンパク質表面構造のエピトープ様領域への分割・データベース 化」の研究を開始した。
